Team solves birth and migration mysteries of cortex's powerful inhibitors, 'chandelier' cells

Team solves birth and migration mysteries of cortex's powerful inhibitors, 'chandelier' cells

Friday, November 23, 2012

The cerebral cortex of the human brain has been called "the crowning achievement of evolution." Ironically, it is so complex that even our greatest minds and most sophisticated science are only now beginning to understand how it organizes itself in early development, and how its many cell types function together as circuits.

A major step toward this great goal in neuroscience has been taken by a team led by Professor Z. Josh Huang, Ph.D., at Cold Spring Harbor Laboratory (CSHL). Today they publish research for the first time revealing the birth timing and embryonic origin of a critical class of inhibitory brain cells called chandelier cells, and tracing the specific paths they take during early development into the cerebral cortex of the mouse brain.

These temporal and spatial sequences are regarded by Huang as genetically programmed aspects of brain development, accounting for aspects of the brain that are likely identical in every member of a given species including humans. Exceptions to these stereotypical patterns include irregularities caused by gene mutations or protein malfunctions, both of which are now being identified in people with developmental disorders and neuropsychiatric illnesses.

Chandelier cells were first noticed only 40 years ago, and in the intervening years frustratingly little has been learned about them, beyond the fact that they "hang" individually among great crowds of excitatory cells in the cortex called pyramidal neurons, and that their relatively short branches make contact with these excitatory cells. Indeed, a single chandelier cell connects, or "synapses," with as many as 500 pyramidal neurons. Noting this, the great biologist Francis Crick decades ago speculated that chandelier cells exerted some kind of "veto" power over the messages being exchanged by the much more numerous excitatory cells in their vicinity.

Born in a previously undiscovered 'country'

After three years of painstaking work that has involved using new technologies to identify and trace neural cell progenitors in ways not previously possible, and to track them as they migrate to positions in the maturing cortex, Huang and colleagues, including Dr. Hiroki Taniguchi now at the Max Planck Florida Institute, have demonstrated that chandelier cells are born in a previously unrecognized portion of the embryonic brain, which they have named the VGZ (ventral germinal zone).

Huang, who has been on a decade-long quest to develop means of learning much more about the cortex's inhibitory cells (sometimes called "interneurons"), points out that while they are far less numerous than the excitatory pyramidal cells all around them, cells including chandelier cells that inhibit or modulate excitatory-cell messages play an indispensable role in balancing message flow and ultimately in determining the functional organization of excitatory neurons into meaningful groups.

This is all the more intriguing in the case of chandelier cells, Huang explains, because of their distinctive anatomy: one cell that can regulate the messages of 500 others in its vicinity is one that we need to know about if we want to understand how brain circuits work. Unlike other inhibitory cells, chandelier cells are known to connect with excitatory cells at one particular anatomical location, of great significance: a place called the axon initial segment (AIS) ? the spot where a "broadcasting" pyramidal cell generates its transmittable message. To be able to interdict 500 "broadcasters" at this point renders a single chandelier cell a very important player in message propagation and coordination within its locality.

Because of the strategic importance of such cells throughout the cortex, it has been a source of frustration to neuroscientists that they (and other inhibitory cells) have been difficult to classify. Huang has pursued a strategy of following them from their places of birth in the emerging cortex.

Many inhibitory cells come from a large incubator area called the MGE (medial ganglionic eminence); until now, it was not known that most chandelier cells are not born there, and indeed do not emerge until after the MGE has disappeared. Only at this point does the much smaller VGZ form, providing a place where neural precursor cells specifically give rise to chandelier cells.

The team learned that manufacture of a protein encoded by a gene called Nkx2.1 is among the signals marking the birth of a chandelier cell. The gene's action, they found, is also necessary to make the cells. Nkx2.1is a transcription factor, whose expression has previously been linked to the birth of other inhibitory neuronal types. Huang's team observes that it is the timing of Nkx2.1's expression in certain precursors -- following disappearance of the MGE and appearance of the VGZ -- that enabled them to track the birth, specifically, of chandelier cells.

Highly specific migration route and cortical destinations

"In addition to being surprised to discover that chandelier cells are born 'late'?after other inhibitory cells ? in a part of the cortex we didn't know about," says Huang, "our second surprise is that once born, these cells take a very stereotyped route into the cortex and assume very specific positions, in three cortical layers." (Layers 2, 5 and 6). "This leads us to postulate that other specific cortical cell types also have specific migration routes in development."

As Huang points out, his team's new discoveries about chandelier cells have implications for disease research, since it is known that the number and connective density of chandelier cells is diminished in schizophrenia. Associations of the same type have recently been made in epilepsy.

"To know the identity of a cell type in the cortex is in effect to know the intrinsic program that distinguishes it from other cell types. In the broadest terms, we are learning about those aspects of the brain development that make us human. 'Nurture,' or experience, also has a very important role in brain development. Our work helps clarify the 'nature' part of the nature/nurture mystery that has always fascinated us," Huang says.

GM Introduces Sail SPRINGO EV in China

General Motors Co announced the launch of its first China-developed electric car at the Guangzhou Auto Show, which will be sold for RMB 258,000 ($41,400).

The Sail SPRINGO EV was developed by Shanghai GM and the Pan Asia Technical Automotive Center (PATAC), making it the first production electric vehicle created by a Chinese joint venture.

The SPRINGO EV pure electric vehicle meets the highest global electric vehicle standards, using the latest industry battery and electric drive technology.

The Sail SPRINGO EV is well suited for urban environments. Its pure electric propulsion system delivers an exceptional driving experience that rivals traditional gasoline-powered vehicles. The vehicle?s electric motor produces minimal sound, for a distinctly peaceful commute.

Based off a new electric vehicle architecture developed by Shanghai GM in conjunction with PATAC, the Sail SPRINGO EV has an emissions-free propulsion system. Its permanent magnet electric motor is capable of producing 85 kW of power and up to 510 Nm of torque, allowing acceleration from 0 to 100 km/h (0-62 mph) from a standing start in just over 10 seconds. Pushing the Sport button provides a 20 percent boost in acceleration at the beginning and middle of the torque curve compared to pressing the accelerator in a gasoline-powered vehicle. The top speed is 130 km/h (80 mph). Its stability during high-speed cruising and cornering is optimized by a new tailored full-frame chassis with an enhanced stabilizer bar and a finely tuned suspension.

The Sail SPRINGO EV?s battery and electric drive technology were developed based on the most advanced global electric vehicle standards for maximum energy efficiency. The result is an exceptionally long electric range of 130 km (80 miles) under a variety of road conditions and 200 (127 miles) km at a constant speed of 60 km/h (37 mph), which is sufficient to meet the daily needs of most people living in urban areas.

The Sail SPRINGO EV consumes less than 15 kWh for every 100 km, which is less than most electric vehicles currently on the market. The Sail SPRINGO EV costs just RMB 4.6 per 100 km to drive when charged during lower-cost off-peak hours (between 10:00 p.m. and 6:00 a.m.).

The Sail SPRINGO EV is environmental and user friendly, with a host of advanced vehicle telematics. It can be charged using a 3.3-kW charging station or a standard household outlet. It takes approximately seven hours for the battery to be fully charged using a dedicated 220V connection. There are three charging modes available: immediate charging, time delay charging, and time delay and rate charging. This enables customers to choose the charging option that fits their needs.

Vehicle charging can also be managed and monitored remotely using a smart phone mobile app provided by Shanghai OnStar. Owners can use the app to perform a range of functions remotely, such as starting the vehicle?s heater or air conditioner. OnStar?s service system also carries out remote vehicle monitoring and analysis of other vehicle systems, and provides drivers with additional vehicle information such as tire pressure. If abnormal data is detected, the OnStar system will immediately contact the owner to provide safety information and instructions.

Yet another feature allows owners to check energy usage and remaining charge level with a touch of the Energy Leaf button. A color display, multimedia entertainment system and information display center with a 7-inch high-definition dual touchscreen offer drivers and passengers an easy-to-use interactive experience.

A world-class safety design protects Sail SPRINGO EV occupants in the event of a collision. The vehicle passed an extensive array of more than 20 collision tests during its development to verify its safety.

Developed to meet the strictest global electric vehicle safety standards, the Sail SPRINGO EV fully complies with safety requirements for pure electric vehicles in China. Its newly designed front and rear anti-collision beams made from ultra-high strength steel and its strong and comprehensive body structure protect the battery pack in the event of a collision. The battery pack and powertrain have performed normally after rigorous testing in various conditions.

All of Shanghai GM's new energy vehicles, including the Sail SPRINGO EV, will be sold through an exclusive ?Drive to Green? sales network. The first showroom is expected to open in the first half of 2013 in Shanghai. The exclusive sales and aftersales service network will be expanded to key new energy vehicle demo cities based on local market conditions and the development of the new energy vehicle industry and charging infrastructure.

The Sail SPRINGO EV will come with a vehicle warranty of three years or 80,000 km (50,000 miles), as well as a battery warranty of five years or 100,000 km (62,000 miles). Until the ?Drive to Green? sales network is established, Shanghai GM's Buick and Chevrolet service centers will provide Sail SPRINGO EV owners door-to-door service and exclusive steward service for a worry-free driving and maintenance experience.

Drained wetlands give off same amount of greenhouse gases as industry

ScienceDaily (Nov. 23, 2012) ? Drained wetlands in Sweden account for the same amount of greenhouse gas emissions as Swedish industry. This is shown by a summary of research from the University of Gothenburg, Sweden and the Swedish University of Agricultural Sciences.

Forests and agricultural fields on drained previous wetlands make up between five and ten percent of Sweden's surface area. When these wetlands are drained, they become a significant source of greenhouse gas emissions.

"We note that drained wetlands which have been forested or used for agricultural purposes are a significant potential source of greenhouse gases of a magnitude that is at least comparable with the industrial sector's greenhouse gas emissions in Sweden."

Emissions from these drained wetlands can be reduced, but that involves rewetting the land -- resulting in a negative impact on forestry production. According to the researchers, compromises may be necessary.

"As long as wetlands remain wet, only methane is given off," says ?sa Kasimir Klemedtsson from the Department of Earth Sciences at the University of Gothenburg. "However, for more than a hundred years land has been drained for agriculture and forestry, producing large quantities both carbon dioxide and nitrous oxide."

Together with researcher ?rjan Berglund from the Swedish University of Agricultural Sciences, Dr Kasimir Klemedtsson was commissioned by the Swedish Environmental Protection Agency during the summer and autumn of 2012 to compile information about greenhouse gases from drained wetlands.

New rules were introduced at last year's Durban Climate Change Conference with the second Kyoto Protocol phase. These rules include the possibility of reporting wetland drainage or rewetting of drained wetlands. Sweden now faces the choice of whether to include these ahead of the second Kyoto Protocol phase.

5 Critical Content Marketing Mistakes That You Must Avoid ...

Content marketing i.e. creating and distributing content is one of the most effective ways to promote a business online. Businesses and marketers all over the world are using content to attract new customers. However, creating quality content regularly, takes valuable time and resources. It is thus important to do it correctly and get the most out of your efforts.

Content Marketing Mistakes

?

In this article we take a look at five common content marketing mistakes that should not be left unchecked. If you want your business to benefit from content marketing, you will need to avoid these mistakes.

1. Writing For The Wrong Audience

When I first started blogging a few years ago, my content was aimed more at marketers, designers and SEO specialists as opposed to businesses. Although my content performed well and I was building a good following, I finally came to a realisation that I was not targeting the right audience.

One of the main purposes of starting a blog for me was to generate leads and sales from my Web design and Internet marketing business. It thus made sense to target businesses instead of other professionals in my niche. Finally, I had to shift focus to address businesses instead of other designers and marketers. I still write about the same topics, but I use less jargon and have had to adjust my tone to address businesses instead. It took me two years to realise this. You can learn from my experience and define your audience right from the start.

2. Poorly Crafted Headlines

Compelling headlines or titles are vital to the success of your content whether it is a blog post, an eBook or a report. Compelling headlines play a major role in determining the popularity and readership of your content.

Content Marketing Mistakes- Poor Headline

A great headline should:

  • Grab the attention of Readers
  • Generate interest and compel users to read the rest of your content
  • Should be clear and concise and easy to understand
  • Should include keywords

You can take advantage of list posts or instructional content as they are very popular among readers and tend to be share more on social media.

Lets take a simple example.? The same headline ?methods to improve the usability of your website can be better written as ?10 Easy Ways to Improve The Usability of Your Website? which is more compelling and will grab the attention of readers.

3. Focusing On Quantity Over Quality

It is often suggested that a business or web master should publish content every day in order to get more coverage and reach. Publishing daily does have its benefits, particular on blogs. It can improve your search engine rank and drive more visitors towards your website. However it should never be at the cost of quality. Publishing quality content once a week can be equally effective for meeting your marketing objectives.

4. Not Optimising Your Content? Search Engine Optimisation

Optimising your content properly is essential to get the desired results. Both, external SEO factors such as link building and on-page optimisation are important. The importance of good on-page optimisation should not be under-estimated. Good optimisation is a must for all content produced as part of your marketing efforts. It should not be overlooked whether you are creating a blog post, press release or an article.

Content Marketing Mistake - No SEO

Source: SEOMoz. Org

Here are some SEO tips you can use:

  • Choose the right keywords:
  • Use Keywords in Headline
  • Use Keywords in Meta title and description
  • Use keywords in headings and bold text
  • Optimise Image ALT descriptions
  • Use in URL e.g. www.yoursitename.com/keywordphrase.html

5. Syndicating The Same Content Everywhere

It is common for businesses and marketers to distribute content on several sources in order to maximise their reach. For example, a business may post content on its blog, publish articles on article sites, create eBooks and as well as publish documents on document sharing sites. Some businesses make the mistake of publishing the same identical content on several sources. ?Although targeting multiple channels can be very effective, syndicating the exact same content everywhere should be avoided.

What About You?

Do you use content marketing to promote your business? Are you avoiding the common mistakes above? Perhaps you would like to add another suggestion to this list. Please share your response by leaving a comment below.
